- Nikon PCM2000 laser scanning confocal coupled to TE-200 epifluorescence microscope
- Objective lenses: 4x, 10x, 20x, 20x MI, 40x WI, 60x WI, 100x oil. Phase contrast and DIC Optics
- Two fluorescence channels and one transmission detector
- Lasers: Argon Ion (488 and 457 nm), HeNe Green (543nm), HeNe Yellow (594nm)
- Orca-1 (1280x1024, 12bit, 2/4/8 binning, 10MHz readout) Hamamatsu CCD Camera. Excitation and emission filter wheels with shutters and controllers.
- Microinjection device: Eppendorf; Temperature controlled and perfusion chambers to maintain the viability of the cells/tissue (Medical Systems Corp.)
- The Simple PCI software drives the hardware. This software is used for confocal-based and camera-based image acquisition and processing.

- Nikon TE300 epifluorescence microscope
- Four-line argon ion laser (457, 488, 514 nm), HeNe green laser (543 nm), and red diode lasers (637 nm)
- Coherent 10W Verdi pumped high-speed ti:sapphire pulsed multiphoton tunable laser system from 700 to 1000 nm
- The software has various functions, including ROI real time logical operation display, FRAP, physiological measurements, various image analysis functions, reverse scanning to reduce cross talk, confocal and multiphoton imaging using the same specimen etc.
- Two channel external detectors for multiphoton imaging; four (3 fluorescence + one transmission) internal detectors for confocal and multiphoton imaging
Zeiss 510 Meta System
- Carl Zeiss 510 Confocal/Multiphoton/META microscopy system coupled to Axioplan 200M epifluorescence microscope
- 40mW argon-ion (458, 488, 514 nm), DPSS 561 nm, Red HeNe 633 nm laser lines
- Verdi 10W pumped Coherent Ti:sapphire laser with x-wave optics tunable from 700-1000 nm
- Temperature controlled stage and Eppendorf microinjection device
Confocal Imaging in Development Biology
The spatial relationship between Myosin IIB (red) and Xenopus fibrillin (XF-green) proteins during early vertebrate development: XF is found surrounding the somites, while myosin IIB is found in the cortex of the cells comprising the somites (left). In contrast, in the notochord XF is found in a medium-angle fiber-winding pattern (30 to 40 degrees), distinct from the low-angle fiber-winding pattern exhibited by myosin IIB (near 0 degrees, relative to the direction orthogonal to the direction of extension-bottom) (Raymond Keller, Biology, UVA).
